Modélisation basée sur la théorie des budgets d’énergie dynamiques (DEB) de la cinétique de bioaccumulation des toxines paralysantes (PST) par l’huître creuse Crassostrea gigas ArchiMer
Pousse, Emilien.
The oyster Crassostrea gigas is one of the most commercialized molluscs in the world. It is a very cosmopolitan species that has been disseminated by humans and is susceptible to various agents such as pathogens (bacteria, viruses), parasites, xenobiotics and/or toxic micro-algae. As a filter-feeder, it may accumulate phyco- toxins while feeding on harmful algal blooms (HAB), including paralytic shellfish toxins (PST). To protect human health shellfisheries harvesting are closed dur- ing HAB, impacting the aquaculture industry. This PhD work aims to develop a Dynamic Energy Budget (DEB) based model to describe kinetics of PST accumulation and detoxification in C. gigas. Interest of coupling bio-accumulation to DEB model lies in the inclusion of...

Morpho-histological characterization of immature of the bioindicator midge Chironomus sancticaroli Strixino and Strixino (Diptera, Chironomidae) Rev. Bras. entomol.
Richardi,Vinicius Sobrinho; Vicentini,Maiara; Rebechi,Débora; Fávaro,Luis Fernando; Navarro-Silva,Mário Antônio.
ABSTRACTChironomidae immature are used as bioindicators of sediment quality in aquatic ecosystems and ecotoxicological assays. Histological descriptions for this family are outdated and limited and there are no studies with Neotropical species. The aim of this study was to describe the tissue architecture of several organs of the larva of Chironomus sancticaroli. For the description of the histological pattern, the larvae were fixed in Duboscq solution for insects at 56 °C, followed by routine histologic processing, infiltration in paraffin, and the sections were stained with Hematoxylin–Eosin. After examining the slides, the tube digestive, salivary gland, excretory, nervous, endocrine, circulatory, and integumentary systems and fat body were...

Parent-to-offspring transfer of sublethal effects of copper exposure: Metabolic rate and life-history traits of Daphnia RChHN
FERNÁNDEZ-GONZÁLEZ,MAURICIO A; GONZÁLEZ-BARRIENTOS,JAVIER; CARTER,MAURICIO J; RAMOS-JILIBERTO,RODRIGO.
In ecological communities, pollution driven perturbations exert immediate effects on sensitive individuals, but these effects may be transmitted among interacting organisms and spread over the community through several paths. This makes the assessment and prediction of ecological consequences of pollution difficult. The propagation of perturbation effects among organisms can be horizontal among organisms that coexist in space and time, and vertical among organism that belong to different generations. The latter process is poorly understood, in particular in planktonic organisms facing metal pollution. In this study we evaluate the vertical transfer of effects driven by sublethal copper stress on the heartbeat rate, somatic growth and fertility of Daphnia...

Protozoa as test organisms in marine ecotoxicology: luxury or necessity? OMA
Dive, D.; Persoone, G..
A critical examination of the utility of protozoan bioassays in marine as well as in freshwater ecotoxicology implies the answer to three basic questions: why, how and when. Arguments in favor of tests with Protozoa are: protozoans combine all biological mechanisms and functions in one single cell; the physiology and biochemistry of several species has been studied extensively; the generation time of Protozoa is very short in comparison to Metazoa; large numbers of organisms can be produced in a small volume; induction of cryptobiotic stages is possible with some species; Protozoa play a significant role in aquatic ecosystems; test methodologies with very sensitive criteria can be worked out. The two basic approaches for bioassay are <i>in...

Relationship between PAH biotransformation as measured by biliary metabolites and EROD activity, and genotoxicity in juveniles of sole (Solea solea) ArchiMer
Wessel, Nathalie; Santos, Raphael; Menard, Dominique; Le Menach, K.; Buchet, Vincent; Le Bayon, Nicolas; Loizeau, Veronique; Burgeot, Thierry; Budzinski, H.; Akcha, Farida.
Polycylic aromatic hydrocarbons (PAHs) are ubiquitous contaminants in the marine environment Their toxicity is mainly linked to the ability of marine species to biotransform them into reactive metabolites PAHs are thus often detected at trace levels in animal tissues For biomonitoring purposes this findings have two main consequences (i) the determination of the PAH tissue concentration is not suitable for the evaluation of individual exposure to PAHs (ii) It can explain sometimes the lack of correlations obtained with relevant markers of toxicity such as genotoxicity biomarkers The aim of the present study was to better investigate the link between PAH exposure and genotoxicity in marine flatfish During a laboratory experiment juvenile soles were exposed...

The originality of this project consists in the interdisciplinary approach gathering the impact of toxic phytoplankton blooms and metallic pollution on health (behaviour, stress, immunity, genetic) and disease development of the Manila clam, an economieally important species. The Manila clam Ruditapes philippinarum is the second exploited bivalve in the world. It was introduced in Arcachon bay during the 80's and is today exploited by professional fishermen (57 licences). A 50% decrease of the stock was observed during the last five years. Beyond fishing pressure, a PhD thesis identified several diseases as potential threats, like perkinsosis (=Dermo) and brown muscle disease (BMD). The project confirms the settlement of these pathologies with high...

Study of the effect of Aliette on the activity of spinach (Spinacea oleracea L.) chloroplasts Braz. J. Plant Physiol.
Almeida,Pedro; Coutinho,João P.; Rocha,João; Gomes-Laranjo,José.
Intensification of production in modern agriculture is frequently obtained thanks to the increase of pesticide applications and chemical fertilizers. However, the use of these products sometimes causes a reduction of crop productivity. We aimed to evaluate the impact of the fungicide Aliette on in vitro activity of chloroplasts of spinach (Spinacea oleracea L.). Transmembrane potential formation, PSII oxygen evolution and PSII photochemistry were analyzed. Our results showed that the recommended concentration (2.5 g L-1) of Aliette did not impair the photosynthetic apparatus. However, at higher concentrations (> 7.5 g L-1), reductions in thylakoid membrane potential and oxygen evolution were recorded, but with no evidence of photoinhibition.

Test écotoxicologique sur microalgues et lecteur de plaques : mesures d'activités enzymatiques sur cellules in vivo. Application à l'évaluation de la qualité du milieu ArchiMer
Gilbert, Franck; Galgani, Francois.
A new method for the assessment of several contaminant effects on marine microalgae (Tetraselmis suesica, Skeletonema costatum) has been developped. The method is based on the measure of cells esterase activity using a fluorimetric stain, fluorescein diacetate (FDA). The experiments are performed in microplates using a fluorimetric microplate reader. The different steps of the experiments and the data processing are controlled by an original specific software. Application of the system for rapid ecotoxicological tests (determination of LC 50) and for assessment of environment quality by observation of water samples are proposed.

Testing the role of contaminants in depressing avian numbers RChHN
KARASOV,WILLIAM H.; MEYER,MICHAEL W..
Environmental contaminants are ubiquitous and so are often key suspects in cases of lagging wildlife populations. How do we test hypotheses about cause-effect linkages between contaminants and wildlife health? We present three case studies in which different approaches were used to test hypotheses about effects of contaminants on wildlife. The cases involve the possible impacts of (1) polychlorinated biphenyl on Lake Superior bald eagles (Haliaeetus leucocephalus); (2) dioxin on osprey (Pandion halieatus); and (3) methyl mercury on common loons (Gavia immer). The different approaches were dictated by legal, logistic, and financial limitations, but the relative strengths of experimental and mechanistic approaches over correlative approaches is underscored....

The monitoring programme of the ecological and ecotoxicological consequences of the "Erika" oil spill ArchiMer
Laubier, Lucien; Le Moigne, Morgan; Flammarion, Patrick; Thybaud, Eric; Cossa, Daniel.
On December 11, 1999, Erika, an oil tanker flying the Maltese flag and loaded with 31 000 t of heavy fuel oil, was making its way from Dunkirk to Livorno when it encountered difficult weather conditions Southwest of the Brittany coast gale force winds 8 to 9 and 6 mwaves). On the morning of 12th December, the captain broadcast an SOS; his vessel was breaking in two, approximately 30 nautical miles to south of Penmarc'h Point (Fig. 1; Finistère, France). The fore section of the vessel sank during the night of the 12-13 December and the aft section was taken in tow on the morning December 13, but subsequently sank in the early afternoon. Both parts lay in 120 m of water and were located 10 km apart. Approximately, 19 000 t of heavy fuel oil were spilled into...

Over the past decade, a significant increase in the circulation of infectious agents was observed. With the spread and emergence of epizootics, zoonoses, and epidemics, the risks of pandemics became more and more critical. Human and animal health has also been threatened by antimicrobial resistance, environmental pollution, and the development of multifactorial and chronic diseases. This highlighted the increasing globalization of health risks and the importance of the human–animal–ecosystem interface in the evolution and emergence of pathogens. A better knowledge of causes and consequences of certain human activities, lifestyles, and behaviors in ecosystems is crucial for a rigorous interpretation of disease dynamics and to drive public policies. As a...

Transcriptional changes in Crassostrea gigas oyster spat following a parental exposure to the herbicide diuron ArchiMer
Rondon, R.; Akcha, Farida; Alonso, P.; Menard, Dominique; Rouxel, Julien; Montagnani, Caroline; Mitta, Guillaume; Cosseau, C.; Grunau, C..
The Pacific oyster Crassostrea gigas is the main oyster species produced in the world, and a key coastal economic resource in France. High mortalities affect Pacific oysters since 2008 in France and Europe. Their origins have been attributed to a combination of biotic and abiotic factors, underlining the importance of environment quality. The impact of water pollution has been pointed out and one of the pollutants, the genotoxic herbicide diuron, occurs at high concentrations all along the French coasts. Previous work has revealed that a parental exposure to diuron had a strong impact on hatching rates and offspring development even if spats were not exposed to diuron themselves. In this study, we explored for the first time the transcriptional changes...

Utilisation des biomarqueurs pour la caractérisation de l’état écotoxicologique des masses d’eau ArchiMer
Sanchez, Wilfried; Porcher, J.-m..
In the past 25 years, numerous biomarkers have been developed with the objective to apply them to aquatic ecosystem biomonitoring. Recently, the Water Framework Directive (WFD) of the European union specified monitoring programs required to assess the achievement of good chemical and ecological status for all water bodies by 2015. This article reviews the potential of biomarkers for ecotoxicological status assessment in WFD monitoring programmes based on previous national and international experiments.
